abstract An assay method, apparatus, and kit using surface plasmons are provided. (A): a step in which a particle having a ligand immobilized on the surface is brought into contact with a specimen; (b): a conjugate of a ligand obtained by (a) with a ligand and an enzyme that is the same as or different from the ligand. (C): the step of further reacting the particles obtained in (b) with an enzyme fluorescent substrate to produce a fluorescent dye, (d): isolating the fluorescent dye obtained in (c) Step (e): A step of bringing the fluorescent dye obtained in (d) into contact with the thin film surface of the plasmon excitation sensor having at least a transparent flat substrate and a metal thin film formed on one surface of the substrate, (f) The amount of fluorescence emitted from the excited fluorescent dye by irradiating the plasmon excitation sensor obtained in (e) with laser light from the other surface of the substrate not formed with the thin film via a prism Obtained by (g) :( f) Calculating the amount of analyte in the sample from the obtained measurement results.
