abstract In a method for labeling proteins with a fluorescent reagent, a fluorescent dye having a fluorophore coumarin and a metal ligand NTA selectively binds to a histag sequence (target protein) to increase fluorescence. This function has the advantage that reversible labeling can be carried out easily and quickly without the need for washing and removing unreacted fluorescent reagents. However, since coumarin is used as a fluorophore, ultraviolet light is used. There is a problem that it is necessary to be excited by, and the application to living cells is very limited. The present invention relates to a ligand that forms a metal complex and exhibits a specific binding ability to a metal coordinating peptide, and a coordination ability to a central metal in the metal complex formed by the ligand.
