Provided are a novel vector, a DNA vaccine and a gene therapy substance containing the vector, a method for producing the vector and a DNA vaccine containing the vector and the gene therapy substance, and a method for therapeutic use of the vector. (A) (i) a DNA binding domain capable of binding to a specific DNA sequence, (ii) a gene expression cassette of a nuclear anchor protein comprising a functional domain capable of binding to a nuclear component, and (b) a nuclear anchor protein A vector comprising a multimerized DNA that forms a binding site for, and (c) one or more expression cassettes of the DNA sequence of interest. It lacks the papillomavirus origin of replication and lacks a functional origin of replication in mammalian cells. Nuclear anchor proteins include the E2 protein of bovine type 1 papillomavirus or Epstein-Barr virus nuclear antigen 1. Further, a method for expressing a target DNA sequence in a subject.
