abstract | Methods and compositions for identifying novel genes that share a homologous region with a known gene from a target group of the gene of interest are provided. The method consists of systematically designing oligonucleotide primers that are specific for the homologous region within the nucleotide sequence of a target group of a known gene, and continuously performing PCR amplification of nucleic acid material from the target organism. including. The PCR step is intended to identify and amplify nucleic acids containing known genes and novel genes. Nucleic acid molecules containing the known gene are detected and removed by further evaluation by dot blot analysis using oligonucleotide probes specific for the known gene in the target group. Potential novel genes are further subjected to sequence analysis to confirm novelty and biological activity is assessed. The compositions of the present invention comprise novel polynucleotides and variants and fragments thereof, which comprise the novel gene and the polypeptide encoded thereby. |
