abstract | The present invention provides a method of determining whether an agent preferentially inhibits presenilin 1 containing γ-secretase over presenilin 2 containing γ-secretase. The present invention also provides an agent that preferentially inhibits presenilin 1-containing γ-secretase over presenilin 2-containing γ-secretase, a pharmaceutical composition containing such a compound, and a method for treating Alzheimer's disease using such a compound. The present invention further discloses that the N-terminal domains of presenilin 1 and presenilin 2 establish differences in the production of Aβ by PS1-containing γ-secretase and PS2-containing γ-secretase. This discovery identified structural determinants for the differences observed in the production of Aβ by PS1-containing γ-secretase and PS2-containing γ-secretase. Such structural determinants have not been previously identified. The present invention also provides a method for determining whether an agent specifically binds to the N-terminus of PS1. The present invention further provides a method for treating Alzheimer's disease by administering an effective amount of an agent that specifically binds to PS1 and consequently inhibits the activity of PS1. |
