abstract The present invention is directed to a method of identifying ribosomal antimicrobial agents that are selective for microbial ribosomes but not selective for mitochondrial ribosomes and / or cytoplasmic ribosomes. Specifically, the method comprises (i) within a bacterial strain having a microbial ribosome, and (ii) within a bacterial strain having a chimeric mitochondrial bacterial ribosome, and / or (iii) having a chimeric cytoplasmic bacterial ribosome. It is intended for assays that compare the interaction of ribosomal antimicrobial candidates within bacterial strains. In a further aspect, the present invention relates to bacterial strains having microbial ribosomes and chimeric forms to identify ribosomal antimicrobial agents that are selective for microbial ribosomes but not selective for mitochondrial ribosomes and / or cytoplasmic ribosomes. It also relates to the use of bacterial strains having mitochondrial bacterial ribosomes and / or bacterial strains having chimeric cytoplasmic bacterial ribosomes. Furthermore, one or more of the bacterial strains (i) to (iii) above can be replaced by a functionally equivalent cell-free biological system.
