abstract | Provided is a substrate of the invention comprising a substrate compound represented by the formula: A- (B 1 -B 2 -B 3 ), wherein A is a sugar moiety, B 1 is the moiety A, and A linker moiety that allows conjugation to the rest of the substrate structure, and B 2 is permanently charged, such as a quaternary ammonium group, for increased proton affinity and ionization efficiency for analysis by mass spectrometry. B 3 has various carbon chain lengths that give specificity to the target enzyme. The present invention also provides a method for detecting lysosomal storage diseases by contacting a sample of the present invention with a sample together with an internal standard, wherein the internal standard is isotopically-labeled. It is an analog of a product obtained by cleavage when a target enzyme is allowed to act on the substrate. |
