abstract | The present invention discloses a novel method for the preparation of recombinant human proteins expressed in human cells. In particular, the present invention relates to a novel process for preparing human recombinant thrombin and human recombinant fibrinogen. Furthermore, the methods of the present invention employ serum-free culture conditions, thereby providing recombinant human proteins expressed in human cells that are more safe for patients when used in human medical treatments. Furthermore, the immunogenic response to recombinant human proteins expressed in human cells can be lower. Although human recombinant thrombin is expressed in the human fetal kidney 293 cell line, the protein can be prepared by two different processes: one starting from point mutant prothrombin with gla and kringle 1 and 2 domains. These domains are maintained during the preparation process; the other starts with prothrombin (non-mutated) and undergoes prethrombin with the HPC4-kringle 2 domain and then makes point mutations to this prethrombin. Human recombinant fibrinogen is expressed in the human fetal kidney 293 cell line. |
