abstract | A method for synthesizing salacinol, its stereoisomers, and analogs, homologues, and other derivatives thereof that are potentially useful as glycosidase inhibitors. The compound of this invention should just have general formula (I) or (II). The synthetic scheme involves reacting a cyclic sulfate with a 5-membered sugar containing a heteroatom (X). The heteroatom preferably consists of sulfur, selenium or nitrogen. Cyclic sulfate esters and cyclic sugar reagents can be readily prepared from carbohydrate precursors such as D-glucose, L-glucose, D-xylose and L-xylose. The target compound is prepared by opening a cyclic sulfate by nucleophilic attack of a 5-membered sugar heteroatom. The resulting heterocyclic compound has a stable internal salt structure composed of a heteroatom cation and a sulfate anion. According to this synthesis scheme, side reactions are limited, and various stereoisomers of the target compound can be obtained in moderate to good yields. |
