abstract | The present invention relates to the provision of in vitro protein synthesis systems and methods for producing solubilized forms of membrane proteins. In some aspects, the invention provides a method of synthesizing a protein using an in vitro protein synthesis system that includes an apolipoprotein, thereby improving the yield of soluble protein over the absence of apolipoprotein. Apolipoproteins useful for the present invention include natural apolipoproteins (including mutated sequences of wild type apolipoproteins) and modified apolipoproteins. The apolipoprotein can be provided to the in vitro protein synthesis system in a state associated with lipid or in a state not associated with lipid. The invention also provides compositions and kits for protein synthesis in solubilized forms, the compositions and kits comprising a cell extract for protein translation and one or more apolipoprotein biomolecules. |
