abstract | The present invention provides compositions, kits, assemblies, libraries, arrays, and high-throughput methods for characterizing large scale structures and functions of gene expression regulatory elements in the genome of an organism, particularly the human genome. In one aspect of the invention, each expression construct varies in a library, a nucleic acid segment operably linked to a reporter sequence in an expression vector such that expression of the reporter sequence is under transcriptional control of the nucleic acid segment. Providing an array of expression constructs comprising nucleic acid segments having a diversity of at least 50; The nucleic acid segment can be a huge library of gene expression regulatory elements such as transcriptional promoters.
