abstract | The present application relates to a method of performing a bisulfite reaction to determine methylation sites in nucleic acids, ie, methylated cytosine and unmethylated cytosine. In this method, the nucleic acid is bisulfite treated and binds to the solid phase when an alkaline solution is added for desulfonation and elution of the nucleic acid from the solid phase. The nucleic acid can be amplified in a further step. The solid phase is preferably a material comprising glass or silica, more preferably a glass fleece, a glass film, or magnetic glass particles. Further disclosed are several uses of alkaline solutions for bisulfite treatment, and kits containing bisulfite reagents, solid phases and alkaline solutions. |
