abstract | The present teachings provide methods, compositions, and kits for detecting microRNA (miRNA). In some embodiments, the miRNA is quantified from formalin-fixed paraffin-embedded tissue in which messenger RNA is degraded. The present teachings take advantage of the observation that most miRNAs are protected from degradation as a result of their association with RISC in vivo. Thus, a novel method for studying nucleic acids in stored tissue containing degraded messenger RNA is provided, where miRNA protected with RISC is released and analyzed. |
