abstract |
The present invention relates to a filamentous phage display method in which a polypeptide of interest displayed on a phage particle is translocated simultaneously with translation across the cell membrane of Gram-negative bacteria based on the signal recognition particle pathway. This method is useful for cDNA libraries and other combinatorials, particularly those derived from polypeptides that are known to be difficult to display on phage, and very quickly folded and stable protein scaffolds. It is particularly suitable for library proteins. The present invention further comprises a phage useful in the above methods comprising a genetic construct encoding a fusion polypeptide comprising a polypeptide to be displayed on a phage particle and an N-terminal signal sequence that facilitates translocation simultaneously with translation or The present invention relates to a phagemid vector. |
