abstract Kind Code: A1 To provide a selective and sensitive nucleic acid detection method and a reliable large-scale screening method for a large number of genomic mutations in heterogeneous biological samples. The present invention provides a mutation detection method having high sensitivity and high selectivity. In a general embodiment, the present invention includes a single base extension reaction that repeats at least once. The methods of the invention are useful in detecting and identifying the presence of microorganisms that cause genetic mutations or diseases in heterogeneous biological samples.
