abstract | A system for enumeration of cells in a fluid by image cytometry for the evaluation of target populations such as leukocyte subpopulations in different body fluids or bacterial contamination in environmental samples, foods, body fluids. explain. The fluorescently labeled target cells are linked to magnetic particles or beads. In one embodiment, a small permanent magnet is inserted directly into the chamber containing the labeled cells. The magnet is coated with PDMS silicone rubber to provide a smooth, flat surface that images on a single focal plane. The magnet is removed from the sample, illuminated with fluorescence, and the light emitted by the target cells is captured by a CCD camera. In another embodiment, the target cells are aligned along a single imaging plane within the sample solution by a floater having a permanent magnet. Image analysis can be performed with a novel algorithm for counting cells on the surface that reflects the target cell concentration of the original sample. [Selection] Figure 1 |
