Disclosed are compounds that inhibit caspases that cause cell apoptosis and inflammation, and pharmaceutical compositions thereof. A compound of formula (I) wherein R 1 , R 2 , R 3 , R 4 , and R 5 represent specific substituents. In addition, pharmaceutical compositions and methods of using such compositions to treat caspase-mediated diseases, as well as processes for preparing these compounds are also provided.
