abstract | [PROBLEMS] To provide a pretreatment method for gene detection of acid-fast bacteria contained in sputum, which is quick, accurate, high yield, simple, and without causing droplet scattering or contamination in a closed system. A pretreatment method capable of safely isolating mycobacteria from sputum is provided. In addition, the present invention provides a method for detecting a mycobacterial gene, wherein the acid-fast bacterium is treated by the pretreatment method of the present invention to extract a gene and the gene is specifically amplified and detected. A pretreatment method for detecting a gene of acid-fast bacteria contained in cocoons, wherein a solubilizing / miscellaneous bacteria removal treatment agent is added to the cocoon, and then the acid is directly removed without removing the treatment agent. A pretreatment method for detecting a mycobacterial gene, which comprises binding and collecting a bacterium by a solid support or a non-specific ligand immobilized on the solid support.
