titleOfInvention Cell processing method for simultaneously detecting and evaluating localization and function of cell membrane proteins
abstract It is possible to simultaneously detect and evaluate the location and function of a cell membrane protein that induces an increase in intracellular Ca 2+ concentration upon activation, and to screen for substances that promote or suppress the activity of the cell membrane protein. A cell processing method is provided. (1) Cell membrane protein A that induces an increase in intracellular Ca 2+ concentration fluorescently labeled with a fluorescent organic compound or fluorescent protein, or a cell membrane that induces an increase in intracellular Ca 2+ concentration fluorescently labeled with a fluorescent protein Gene A ′ encoding protein A is prepared. (2) A fluorescent probe protein B for detecting Ca 2+ that increases in concentration in the cell with the activation of the above-mentioned cell membrane protein A, or a gene B ′ encoding the same is prepared. (3) C is prepared by adding a nuclear translocation signal peptide to B, or C ′ is prepared by adding a gene encoding the nuclear translocation signal peptide to B ′. (4) A combination of either A + C or A ′ + C ′ is introduced or expressed in the same cell. [Selection] Figure 1
