abstract Disclosed is a lysis reagent composition that does not contain cyanide, and a method of use for measuring total hemoglobin concentration and leukocytes in a blood sample. The lytic reagent composition includes a quaternary ammonium surfactant, which lyses red blood cells to liberate hemoglobin and ligand, forming a stable chromogen with hemoglobin. The lytic reagent composition has a pH in the range of 3-10. The lytic reagent composition can also include a second quaternary ammonium surfactant.
