abstract |
CD4 + regulatory T (Treg) cells greatly suppress the host immune response, thus protecting against autoimmune diseases while limiting the desired immune response (eg, anti-tumor immunity). Synthesized phosphorothioate-protected guanosine-containing oligonucleotides can directly reverse the inhibitory activity of Treg cells without affecting dendritic cells. This effect appears to be transmitted by signaling through Toll-like receptor (TLR) 8 in Treg cells and the involvement of MyD88 and IRAK4 molecules. Stimulation of Treg cells with natural ligands against human TLR8 reproduced the action of synthesized guanosine-containing oligonucleotides. |
