abstract | Compositions, methods, and kits for in vitro synthesis systems of biomolecules such as polypeptides are provided in the present invention. Cell extracts that provide improved yields of soluble protein using in vitro protein synthesis methods are provided. The present invention also includes a method for adding a feed solution comprising an amino acid and an energy source to an ongoing in vitro synthesis system to obtain a high protein yield. The invention also includes the use of a high yield in vitro synthesis system to produce large amounts of protein with labeled amino acids incorporated for analysis by methods such as NMR. The present invention further includes vectors for improving protein production from template nucleic acids using an in vitro synthesis system. |
