titleOfInvention | Two-dimensional strand number-dependent and length-dependent separation of nucleic acid fragments |
abstract | A method is provided for separating single and double stranded nucleic acid molecules based on their number and length. The method is based on a novel two-dimensional gel electrophoresis technique, in which a sample of nucleic acid molecules is loaded into a gel electrophoresis apparatus and the sample is passed through a gel matrix in a first dimension under a first set of predetermined electrophoresis conditions. Electrophoresis of the gel matrix in a second dimension under a second set of electrophoresis conditions, thereby separating a population of single-stranded and double-stranded nucleic acids, wherein The first and second electrophoresis conditions are different so that in one dimension the electrophoresis separates the sample molecules based on the number of strands and length, and in the other dimension the electrophoresis is substantially lengthy. Where the differences are established by chemical agents and / or physical parameters that affect the migration rate of the number-dependent electrophoresis of nucleic acids. [Selection] Figure 13 |
