abstract | The present invention provides a method for assaying myeloperoxidase activity. In this assay, a sample containing myeloperoxidase or a sample presumed to contain myeloperoxidase is contacted with a substrate containing serine, hydrogen peroxide, and halide. If myeloperoxidase is present in the sample, serine is converted to glycol aldehyde, which is further converted to glycolate by glycol aldehyde converting enzyme (eg, glycol aldehyde dehydrogenase) (1.2.1.21). Converted. The method then utilizes a circulating reaction system between glycolate and glyoxylate to provide a detectable signal corresponding to this myeloperoxidase activity. Kits for assaying myeloperoxidase based on the same principle are also provided. |
