abstract |
A mutant hydrolase optionally fused to the protein of interest is provided. This mutant hydrolase can form a more stable bond with the substrate of the corresponding non-mutant (wild type) hydrolase than the bond formed between the wild type hydrolase and the substrate, and compared to the wild type hydrolase. Containing two amino acid substitutions. Hydrolase substrates containing one or more functional groups are also provided, along with methods of using the mutant hydrolases and substrates of the present invention. Also provided are fusion proteins capable of forming stable bonds with the substrate and cells expressing the fusion proteins. |
