abstract Disclosed are solid phase materials for nucleic acid binding and methods relating to their use. The material features a cleavable binding moiety that can be cleaved to dissociate the bound nucleic acid. The solid phase material comprises a solid support portion comprising a substrate selected from silica, glass, an insoluble synthetic polymer, and an insoluble polysaccharide that binds to a nucleic acid binding portion for attracting and binding nucleic acids, said nucleic acid binding portion (NAB) is bound to the solid support portion by a cleavable binding moiety. Preferably, the nucleic acid binding moiety comprises a tertiary or quaternary onium group. The material is a fine particle, fiber, bead, membrane, test tube or microwell and may include a magnetic core. Nucleic acid binding methods using a cleavable solid support are disclosed for use in nucleic acid isolation or purification methods.
