An isolated modified fibronectin binding protein (Fnb) of S. aureus, comprising S. aureus Fnb having at least one mutation in an amino acid selected from residues corresponding to Gnb103, Gln105, Lys157, Lys503, Lys620, Lys702, Lys762, Gln783, and Gln830 of FnbA of Aureus strain ATCC 49525 is described. By substituting these reactive residues in the fibronectin binding protein, the protein is less able to covalently crosslink fibronectin and fibrin than wild-type Fnb. The modified fibronectin binding protein is S. cerevisiae. An immunogenic composition that effectively interferes with the adherence of aureus to fibronectin and fibrin, and thus includes such modified Fnb, exhibits improved immunogenicity and is safer to use. |
