abstract |
The present invention relates to a type of controlled release multilayer methylphenidate pellet that does not need to be combined with another type of pellet. It is possible to maintain a therapeutic concentration in plasma for 12 hours with once daily dosing, avoiding repeated dosing throughout the day. Controlled release multilayer pellets are an inert core, a first layer containing methylphenidate and an acid buffer system, a protective layer, and a layer of ethylcellulose that functions to control the sustained release of most methylphenidate. And a second methylphenidate layer responsible for immediate release within 1 hour of administration. The weight ratio between methylphenidate and ethylcellulose present in the first active layer is 1.40: 1 to 1.90: 1. The multi-layer pellet may have an outer coating to protect it from erosion during processing. |
