abstract | Provided is a method for easily measuring the content of methylated DNA in a target DNA region possessed by genomic DNA contained in a biological specimen. [MEANS FOR SOLVING PROBLEMS] (1) Obtaining methylated single-stranded DNA from a genomic DNA-derived DNA sample contained in a biological specimen, and binding the single-stranded DNA to an immobilized methylated DNA antibody. A first step of selecting single-stranded DNA, and (2) a second step of digesting the single-stranded DNA selected in the first step with a methylation sensitive restriction enzyme capable of digesting at least one kind of single-stranded DNA. And (3) as a pre-process of each of the following steps, the single-stranded DNA, which is an undigested product obtained in the second step, is separated from the immobilized methylated DNA antibody into a single-stranded state.
