abstract It is possible to prepare a hydrogenase enzyme solution in a complete state in which degradation of small subunits does not occur at a high recovery rate, and to easily obtain an enzyme solution having a high activity per unit weight. A method for isolating and purifying hydrogenase is provided. Also provided are a hydrogenase enzyme solution that is homogeneous and has a high activity per enzyme unit weight, and a fuel cell using the hydrogenase enzyme solution as a catalyst component. When isolating and purifying hydrogenase using column chromatography, a sample containing hydrogenase is loaded onto the column using only a neutral to weakly acidic buffer and the hydrogenase is eluted from the column. A method for isolating and purifying hydrogenase, a hydrogenase enzyme solution that is isolated and purified, comprising a highly homogeneous hydrogenase that does not contain degradation products, and the hydrogenase enzyme A fuel cell comprising an electrode catalyst produced using a solution.
