Preparation of a stable formulation of lipid-nucleic acid complex effective for in vivo delivery |
abstract |
The present invention provides lipid-nucleic acid complexes with increased storage stability and high transfection activity in vivo after intravenous injection, and methods for preparing such complexes. This method generally involves contacting a nucleic acid with an organic polycation to produce a condensed nucleic acid, and then combining the condensed nucleic acid with a lipid having an amphiphilic cationic lipid to produce a lipid-nucleic acid complex. Including doing. This complex can be further stabilized by adding a hydrophilic polymer to a hydrophobic side chain. This complex can also be specific to a particular cell by incorporating it into a target moiety such as a Fab fragment attached to a hydrophobic polymer. [Selection figure] None |
