An expression system is provided that provides a heterologous protein that is expressed by a non-native host organism but has biological activity and / or structure similar to that of a natural protein. Disclosed are vectors, expression hosts, and methods for expressing heterologous proteins. Expression systems include co-expression of a protein factor that can catalyze disulfide bond formation and a desired heterologous protein. Expression systems using yeast cells as preferred hosts, protein disulfide isomerase (PDI) and thioredoxin (TRX) as preferred examples of protein factors, and HCV-E2 715 envelope glycoprotein and human FIGF as preferred examples of heterologous proteins are shown.
