PROBLEM TO BE SOLVED: To provide a method for measuring cholesterol (LDL) in low density lipoprotein in a biological sample such as serum and plasma, and a reagent thereof. In a method for measuring cholesterol in low-density lipoprotein by enzymatic reaction, pH 7.6-9. Containing cholesterol esterase, cholesterol oxidase, and a substance that inhibits reactivity to these low-density lipoproteins in the first reaction. After preferentially erasing high density lipoprotein, ultra-low density lipoprotein and cholesterol in chylomicron in the buffer of 0, in the second reaction, the low-density lipoproteins of both enzymes in the first reaction are removed.
