abstract | A novel raw starch-degrading enzyme produced by an earthworm is provided. The lyophilized powder of earthworm is suspended in a buffer solution and centrifuged, and then the supernatant is recovered. After adding a protease inhibitor to the supernatant and ultracentrifugating, the supernatant obtained is saturated to 35%. Fraction having two raw starch degrading enzyme activities by adding ammonium sulfate and dissolving the resulting precipitate in a small amount of buffer solution and subjecting it to anion exchange chromatography using the degradation activity on starch or raw starch as an index. (The active fraction eluted first was named as raw starch degrading enzyme I, and the active fraction eluted later was named as raw starch degrading enzyme II). These fractions were subjected to gel filtration chromatography and hydrophobic chromatography, respectively, and the active fractions were collected to obtain uniform raw starch-degrading enzyme I and raw starch-degrading enzyme II produced by the earthworm.
