abstract | Disclosed is a DNA segment encoding a recognition domain and a cleavage domain of FokI restriction endonuclease. The 41 kDa N-terminal fragment constitutes the recognition domain of the FokI restriction endonuclease, and the 41 kDa C-terminal fragment constitutes the nuclease domain of the FokI restriction endonuclease. A hybrid restriction enzyme (Ubx-F N ) containing the nuclease domain of the Fok I restriction endonuclease by linking the ultrabithoracic Ubx homeodomain to the open domain (F N ) of Fok1.
