abstract | The methods and apparatus disclosed herein relate to nucleic acid sequencing by enhanced Raman spectroscopy. In certain embodiments of the invention, the nucleotide is covalently linked to the Raman label before being incorporated into the nucleic acid. In other embodiments, unlabeled nucleic acids are used. When the nucleic acid is treated with exonuclease, labeled or unlabeled nucleotides are released and detected by Raman spectroscopy. In another embodiment of the invention, the nucleotide released from the nucleic acid by exonuclease treatment is covalently crosslinked to the nanoparticle, surface enhanced Raman spectroscopy (SERS), surface enhanced resonance Raman spectroscopy (SERRS), and / or Detected by coherent anti-Stokes Raman spectroscopy (CARS).
