abstract | Geldanamycin derivatives, which block the uPA-plasmin network at femtomolar concentrations and inhibit the growth and invasion of glioblastoma cells and other tumors, have the potential to be highly active anticancer agents. Disclosed are GA and various 17-amino-17-demethoxygeldanamycin derivatives that block Met tyrosine kinase receptor-dependent uPA activation mediated by HGF / SF at the fM level. The inhibitory activity of the compounds tested is incompatible with the already known ability of this class of drugs to bind hsp90, which is a new indication for events mediated by HGF / SF in tumor development. Indicates that the target is present. Disclosed are methods for using such compounds to inhibit the activity of cancer cells and to treat tumors. |
