abstract |
The present invention provides an emulsion based method for preparing controlled release microspheres for delivery of active compounds. The method includes the preparation of an emulsion comprising an aqueous dispersed phase comprising a polymer capable of forming a hydrogel, a bioactive protein, and water and substantially free of insoluble aggregates of the bioactive protein. Subsequently, the polymer is physically or chemically cross-linked to form a hydrogel. The present invention further provides active protein-filled hydrogel microspheres prepared by the method and substantially free of insoluble aggregates of the active protein. The microspheres exhibit controlled release with a release profile that is significantly improved over the release profile of known hydrogel microspheres. The microspheres can be used to deliver therapeutic or diagnostic proteins by injection. [Selection] Figure 5 |
