Highly purified preparations of TFPI or TFPI analogs can generally be prepared using a method comprising the following steps: (1) E. Expression of TFPI or TFPI analog in E. coli, (2) Refraction isolation, (3) Refraction dissolution, and refolding of expressed TFPI or TFPI analog, (4) SP-Sepharose high flow rate (FF) Chromatography, (5) primary concentration step and diafiltration step, (6) Q-Sepharose high-speed (HP) chromatography, (7) Butyl hydrophobic interaction chromatography (HIC), (8) SP-Sepharose HP Chromatography, and (9) Secondary concentration step / diafiltration step. |
