abstract |
The present invention provides reagents and methods for accurately quantifying proteins in complex biological samples. To perform quantification, a peptide combo is added to the sample, which is essentially a collection of synthetic reference peptides. The mass difference in the synthetic reference peptide is small compared to a biological reference peptide obtained by digesting a protein present in the sample. A reference peptide and a synthetic reference peptide are selected and the identity and exact amount of the reference peptide are determined by mass spectrum. This method can be used in high throughput assays to analyze the proteome. |
