titleOfInvention | Protein fractionation using high performance tangential flow filtration |
abstract | A process and apparatus are provided for separating a molecule from a mixture containing the molecule of interest. This process involves subjecting the mixture to an improved method of high performance tangential flow filtration (HPTFF). Using the HPTFF of the present invention, various feed streams were purified and treated to remove the target molecule based on both the size and charge of the target molecule. According to a preferred embodiment, the transgenic milk feed stream is stabilized and particulate matter such as fat, casein micelles and bacteria is removed. The HPTFF method used in the present invention utilizes optimized process parameters including temperature, transmembrane pressure, molecular charge, molecular size, crossflow rate, and milk concentration. Cleaning and storage procedures have also been developed to ensure long membrane life. A sterile filtration process has also been developed to remove any bacteria remaining in the purified transgenic milk feed stream. |
