abstract | Lipopolysaccharide (LPS, endotoxin) is released along with the protein of interest during recombinant protein production from gram-negative bacteria. In many instances, LPS is purified along with the protein of interest by specific or non-specific protein-ILPS interactions. The inventors examined the ability of alkanediol to separate LPS from protein-LPS complexes while immobilizing the complex to an anion or cation exchange chromatography medium. Alkanediol provides a safer alternative to the use of other organics such as alcohol or acetonitrile because it is less toxic and less flammable. Furthermore, they are cheaper in cost than many detergents that have been used for such purposes. LPS removal efficiency increased as the alkane chain lengthened. 1,2-alkanediol was more effective than terminal alkanediol in separating LPS from protein LPS complexes. |
