abstract The present invention provides allosterically regulatable polynucleotides that have target-dependent circularization and topological binding ability to target nucleic acid molecules. The polynucleotides of the invention include target binding sequences and regulatory elements (which prevent circularization when target binding is not present). A polynucleotide can include a catalytic domain that advances catalytic circularization when the target binding sequence of the polynucleotide is bound to a target. Topologically bound polynucleotides can be used to detect target molecules or to inhibit transcription or translation of a target.
