abstract A mutant hydrolase optionally fused to a protein of interest is provided. The mutant hydrolase is a bond with a substrate of a corresponding non-mutant (wild-type) hydrolase that is more stable than the bond formed between the wild-type hydrolase and the substrate. Can be formed. A hydrolase substrate comprising one or more functional groups is also provided, as is the method using the mutant hydrolase and substrate of the present invention. Also provided are fusion proteins that can form stable bonds with the substrate and cells that express the fusion proteins.
