abstract | Methods and compositions are provided for enhancing the specificity and sensitivity of amplification reaction mixtures. The composition according to the invention provides a reduction in primer aggregate formation during the amplification reaction. The reaction mix contains dNTPs, some of which are replaced by unusual nucleotides, such as dUTP. The concentration of non-conventional nucleotides is typically 10% to 50% of the concentration of one of the standard dNTPs. In some compositions, this unusual nucleotide is dUTP replacing about 10% to about 50% of dTTP in the dNTP mix. |
