Synthetic peptides based on the native sequence of HIV gp41 HR2 are provided provided that (a) a helix-promoting amino acid, or (b) a helix-promoting amino acid and a charged amino acid (to form an ion pair in the synthetic peptide). A synthetic peptide having a plurality of amino acid substitutions, and a synthetic peptide that is unpredictably improved compared to a peptide having an amino acid sequence that does not contain a plurality of amino acid substitutions. Shows activity. Further provided are polynucleotides that encode the synthetic peptides, and methods of using the synthetic peptides as compositions for blocking the spread of HIV to target cells or as a composition for blocking the spread of HIV to target cells. |
