abstract | Methods and kits are provided for detecting the presence or absence of a target nucleic acid sequence in a sample. The methods and kits include the use of metal nanoparticles and electrostatic interactions between the metal nanoparticles and the nucleic acid molecule. The method relies on the differential interaction of ss-nucleic acid and ds-nucleic acid with metal nanoparticles. The colorimetric detection approach takes advantage of the ability of ss-nucleic acids electrostatically associated with metal nanoparticles in a colloidal suspension to stabilize them against aggregation. Fluorescence approaches involving tagged ss-oligonucleotide probes convert the differential adsorption of ss-nucleic acids onto metal nanoparticles to the differential quenching of fluorescent tags on probes that did not hybridize to the target. |
