abstract | According to the present invention, an enzyme responsible for prodrug activation is identified and used for identification of candidate compounds as prodrugs. The invention encompasses methods of identifying candidate compounds as suitable prodrugs and methods of screening candidate compounds for suitability as therapeutic agents. The above identifying method includes the steps of (a) providing a candidate compound having an esterified phosphonate group or an esterified carboxyl group; (b) contacting the candidate compound with an extract containing GS-9005 ester hydrolase A. And (c) at least one of the one or more metabolic compounds has a phosphonic acid group instead of the esterified phosphonate group of the candidate compound, or of the candidate compound Identifying a candidate prodrug as a suitable prodrug when it has a carboxylic acid group instead of an esterified carboxyl group. |
