abstract |
The methods and apparatus disclosed herein relate to characterization of nucleic acids by enhanced Raman spectroscopy. In certain embodiments of the invention, exonuclease treatment of the nucleic acid results in the release of nucleotides. The nucleotide can pass from the reaction chamber through the microfluidic channel and into the nanochannel or microchannel. This nanochannel or microchannel may be filled with a nanoparticle assembly that includes hot spots for Raman detection. As the nucleotides pass through the nanoparticle hotspot, they can be detected by Raman spectroscopy. Identification of the sequence of nucleotides released from a nucleic acid is used to characterize the nucleic acid, for example, by sequencing or identifying the nucleic acid. Another aspect of the invention relates to an apparatus for nucleic acid sequencing. |
