abstract |
Methods are provided for detecting the formation of complexes of molecules (particularly proteins) in a sample such as a cell or tissue lysate. In one embodiment, a cleaving probe specific for a first protein in the complex and one or more binding compounds specific for one or more second proteins in the complex are provided. Upon binding, the cleavable probe is induced to produce an active species such as singlet oxygen, which cleaves the molecular tag attached to the binding compound only within the local region of the cleavable probe. The released molecular tags are separated from the assay mixture and from each other to obtain a measurement that is related to the number and type of proteins present in the complex. |
